Ribonuclease H also known as RNase H is an enzyme that remove the RNA primers on the lagging strand during DNA replication. RNA primers are replaced with nucleotides between the Okazaki fragments[1][2]. During DNA replication, the synthesis of a new strand of DNA on the lagging strand is discontinuously due to the formation of Okazaki Fragment. RNase H will cleave the RNA primers from the Okazaki fragment and replace it with DNA. DNA ligase can from phosphodiester bond between the nucleotides added and the Okazaki fragment. This result in the formation of a complete continuous strand of new DNA.
