Maximum Velocity (Vmax) occurs when all enzymes are working at they're maximum speed at high substrate concentration. The enzymes are said to saturated by the substrate; increasing the substrate concentration will not increase the velocity of the reaction. Maximum velocity can be determined from a plot of rate of product formation (initial velocity) against substrate concentration.
