 Coomassie Brilliant Blue G-250 is a triphenylmethane which is commonly used as a dye to visuallise protein bands during PCR. 

Methology of staining the polyacrylamide with the dye is as follows

  1. If the gel contains SDS then wash it 3 times for 5 minuetes each in deionised water.
  2. Cover the gel in Coosassie Brilliant Blue G-250 dye for 1 hour, the proteins bands will begin to appear within 5 minuetes however will be at there peak of intensity within 1 hour.
  3. Repeat stage 1 to intesify the protein bands.
