Pfu polymerase is an enzyme isolated from the organism Pyrococcus furiosus, a hyperthermophilic archaeon. This organism can survive temperatures of up to one hundred degrees celcius, therefore is very apt for use in the PCR reaction, as, unlike DNA polymerase, it can survive and function at the high temperatures the process operates at. It may be preferred to the use of Taq polymerase (another thermostable enzyme) as pfu has 3'-5' proof reading activity, improving the accuracy of copying DNA. =
